Abstrak

Kajian anatomi perbandingan daun dilakukan ke atas daun Sauropus androygnus yang dijangkiti virus Sauropus yellowing (SaYV). Objektif kajian ialah untuk mengenal pasti ciri anatomi daun yang dijangkiti virus SaYV dan daun sihat (kawalan) serta menentukan ciri anatomi yang berubah dengan jangkitan virus SaYV. Kajian melibatkan keratan rentas lamina, petiol dan tepi daun menggunakan mikrotom gelongsor, teknik penjernihan untuk peruratan dan siatan epidermis untuk epidermis adaksial dan abaksial daun. Hasil kajian menunjukkan pengurangan peratusan pigmen klorofil pada lamina dan tepi daun, hiperplasia tisu vaskular pada tulang daun dan petiol, hiperplasia sel kolenkima dan pertambahan kepadatan hablur drus pada berkas vaskular lamina, tulang daun dan tisu mesofil. Terdapat kerosakan pada urat sekunder dan tertier daun akibat jangkitan virus. Hasil cerapan pada epidermis abaksial daun dijangkiti menunjukkan ketumpatan dan indeks stoma (SI) yang rendah berbanding daun sihat. Kajian ini membuktikan berlaku pengubahsuaian ciri anatomi S. androygnus akibat jangkitan virus SaYV sebagai respons terhadap jangkitan virus. Hasil kajian dapat memberi manfaat kepada pengusaha pertanian dan boleh menjadi rujukan dalam pengurusan penyakit pertanian terutama untuk kawalanpenyakit pada awal jangkitan virus.

Abstract

A comparative leaf anatomy study was performed on Sauropus androygnus leaves infected with Sauropus yellowing virus (SaYV). The objective of the study was to identify the anatomical features of leaves infected with SaYV virus and healthy leaves (control), as well as to determine the anatomical features that change with SaYV virus infection. The study involved cross-sectioning of the lamina, petiole, and leaf margin using a sliding microtome, clearing techniques for leaf venation, and epidermal peels for the adaxial and abaxial epidermis. The results of the study showed a reduction in the percentage of chlorophyll pigment on the lamina and leaf margin, vascular tissue hyperplasia on the leaf midrib and petiole, collenchyma cell hyperplasia, and an increase in the density of druses on the vascular bundles of the lamina, leaf midrib, and mesophyll tissue. There is damage to secondary and tertiary leaf veins due to virus infection. Observation results on the abaxial epidermis of infected leaves show a low stomatal density and index (SI) compared to healthy leaves. This study proves that there is a modification of the anatomical features of S. androygnus due to SaYV virus infection in response to the virus infection. The results of the study can benefit agricultural operators and can be a reference in the management of agricultural diseases, especially for disease control at the beginning of virus infection.
